Mass Effect 1, slowly realizing the real enemy isnt some rogue spectre but his giant spaceship thats apparently sentient and wants to wipe out all life.
When i first played mass effect 1 it had only just come out, and the first convo you have with this 'guy' gave me goosebumps

Harbinger possessing the collectors in mass effect 2 always terrified me
This stupid fucking bitch could literally spam massive AOE attacks with no reset time and stun-lock you until she got her Ultimate and fucking wiped you out.
To this day, I have not beaten her with any of the fighters I'm good with (Ryu, Sagat, Zangief, Abel, E. Honda)

Tarkus from Dawn of War 2 Chaos Rising.
The traitor can actually by any of your team members, depending on what you do during the campaign, but I find Tarkus's betrayal the most interesting. Basically, he finds a possessed bolter which brainwashes him into thinking that he can use its power to defeat chaos and save the Imperium and all his friends.
When anyone else turns to chaos they all go KILL YOU ALL UNWORTHY SCUM FEEL MY WRATH but Tarkus spends the fight trying to explain his reasoning and asking you to just leave because he doesn't want to have to hurt you. He may not have been able to resist chaos corruption but it was never able to destroy his morals and loyalty, and so it had to twist them around instead.

I would love to see Glados and Handsome Jack get locked in a room together.
The buildup and emotional payout of finally defeating this guy still brings a tear to my eye.
