Restarting won't fix it, seems to be a general flaw of the filemanager that it forgets all thumbnails on external drives the second the drive is gone or the computer is off. Bad hardware doesn't help.

Explorer furgets about thumbnails sometimes fur me too. Butt knot all of them, it still shows one every here and there. Happens completely random and very rarely. I dokn't need to do anything to fox it tho because usually the next time I open Explorer it's all back to normal again
Note if you use an utility that cleans up various caches (such as CCleaner) it could also wipe Thumbnail Cache
However, it's pawsible since you mentioned big folders and external drive Windows just doeskn't manage to write Thumbs.db quickly enough when your browsing stuff so the cache will have to regenerate every time you open the folder
